NAC (N-Acetyl Cysteine) provides cysteine, the rate-limiting precursor to glutathione synthesis. Supplementing cysteine directly raises intracellular glutathione levels across liver, muscle, immune and neural tissue. L-Glycine is the smallest amino acid, serving as the third component of the glutathione tripeptide alongside cysteine and glutamic acid. Glycine supplementation has been shown to correct the age-associated glycine deficit that limits glutathione synthesis even when cysteine is available. Research from the Baylor College of Medicine demonstrated that GlyNac supplementation in older humans corrected glutathione deficiency, reduced oxidative stress, improved mitochondrial function, and improved several biomarkers of aging including insulin sensitivity and muscle strength - results not seen with NAC or glycine alone.
What separates GlyNac from standalone NAC supplementation is glycine co-delivery. NAC raises cysteine availability; without adequate glycine, cysteine cannot be converted to the complete glutathione tripeptide. In younger individuals this is rarely limiting because dietary glycine from collagen-rich foods is typically adequate. The combination of both precursors simultaneously addresses both bottlenecks in the glutathione synthesis pathway.
